The 25-year-old Dutch forward opened the scoring after 31 minutes, heading home from close range shortly after teammate Mattia Zaccagni had a penalty saved.
Giovanni Simeone equalised for Napoli soon afterwards, but Noslin put Lazio back in front four minutes before half-time.
Noslin completed his first career hat-trick soon after the interval by expertly heading a cross into the far corner.
Napoli, who started with several key players on the bench, including Khvicha Kvaratskhelia, Romelu Lukaku and Scott McTominay, were unable to mount a comeback and Lazio eased into the quarterfinals.
